半乳糖激酶（英語：Galactokinase）是一種催化α-D-半乳糖磷酸化為半乳糖-1-磷酸的磷酸轉移酶，在此過程中消耗一分子 ATP^[1]。半乳糖激酶催化Leloir途徑（有機體中將α-D-半乳糖轉化為葡萄糖-1-磷酸的代謝途徑）的第二步反應，最早在哺乳動物的肝臟中發現, 後來在酵母、古菌^[2]、植物^[3]^[4]和人類中都有發現^[5]。
